The DP2 is one of the few NIH mechanisms that explicitly rewards ideas too bold for regular study section review. Most ESIs hear about it and assume it's out of reach. In practice, it's underused precisely because the application logic is different — not because the science has to be impossibly transformative.

DP2 reviewers are reading for something different: a specific, testable claim about why the world looks different if you succeed.

The framing that tends to work is more direct about stakes and uncertainty. Something like: 'If this experiment succeeds, the conceptual model for X will need to be revised' or 'This has not been done because no existing technology could do it; ours can, for the first time.' These are claims about transformative potential, not about probability of near-term success. They invite reviewers to evaluate whether the upside is worth the risk, which is the right question for this mechanism.

One practical test: read your closing paragraph of the six-page section and ask whether a reviewer can describe in one sentence what would be different about the field if your aims succeeded. If the answer is vague — 'we will better understand mechanism X' — it's not a DP2 argument yet. If the answer is specific and surprising — 'we would have the first direct evidence that cell type Y drives disease Z, reversing a 20-year assumption' — you're in the right territory. That same test applies to your Specific Aims page. Because the Research Strategy is only six pages, the Aims page cannot defer the transformative argument to the body — the central claim has to land in the first two hundred words. If a panelist reads the Aims and thinks 'interesting but safe,' the score will reflect that before they open anything else.

When to Choose the DP2 Over an R21 or R01

The DP2 occupies a different niche from both the R21 and the standard R01. An R21 is exploratory and short (two years, smaller budget). It's appropriate when you need to establish feasibility before committing to a larger mechanism. A DP2 is appropriate when the idea is mature enough to execute if funded, but too speculative for R01 review because you can't yet produce the preliminary data that a study section would require. If you're still testing whether the core assay works, the R21 fits better. If you have a working approach but not the de-risking data a conventional panel demands, the DP2 is worth considering.

An R01 remains the standard first independent award for most faculty, and for most applicants it is still the right move — it aligns with study section culture and with program officer relationships at the institute level. The DP2 makes sense when your science is genuinely novel enough that a conventional panel would ask for data you cannot yet have. If your idea would score well in a regular review, the R01 is usually faster and more predictable than a DP2 application.

Don't assume the DP2 and R01 are mutually exclusive in the same submission cycle. Check the active FOA language and the application count limits. In some cycles applicants have submitted both, but the strategic cost of two full applications is real. Assess whether you can write both well — not both adequately — before committing to that calendar.
